Hepatitis A vaccine is safe and highly effective in adults, with a single dose triggering strong immunity and a second dose boosting protection to 100% across all age groups.

A new clinical investigation has offered encouraging news for adult vaccination: the hepatitis A vaccine is not only safe but highly effective, whether given as a single shot or as part of a two-dose schedule.

More than 1,000 adults, aged 18 and older, took part in the trial. Before vaccination, blood tests revealed that just over half of the participants (52.56%) already had antibodies against the hepatitis A virus. Surprisingly, the lowest natural immunity was seen in people aged 36 to 40 years, suggesting this age group may be particularly vulnerable. All participants received one dose of the hepatitis A vaccine. Those who tested negative for antibodies were then divided into groups, with some given a second dose at different intervals. Researchers tracked antibody levels after each vaccination and monitored side effects for nearly a month.

The results were striking!

  • First dose impact: Within a month of the first shot, the seroconversion rate reached 67.68%, showing that a majority of previously unprotected adults developed immunity quickly.
  • Booster effect: In the group given a second dose after 6 months, both seropositivity and seroconversion hit 100%. Antibody levels also skyrocketed, with a geometric mean concentration (GMC) of 3,602.44 IU/L 1 month after the booster.
  • Consistency across ages: Antibody levels did not vary markedly across different age brackets, underscoring the vaccine’s broad effectiveness.
  • Safety profile: Mild side effects were reported in only 3.85% of subjects after the second dose, and importantly, no serious adverse events were linked to the vaccine.

The study’s interim findings reinforce that the hepatitis A vaccine yields robust protection with just one dose, while a second dose ensures complete and long-lasting immunity.
